Luciferase-tagged Cells

Luciferase-tagged cells are living cells genetically modified to produce luciferase, an enzyme that generates bioluminescence and enables researchers to monitor cell behavior noninvasively. After administration of a compatible luciferin substrate, luciferase catalyzes a light-producing reaction, and the emitted signal can be detected with sensitive imaging systems. In medicine, this approach supports real-time tracking of cell survival, proliferation, migration, and distribution in disease models and during cell-based therapies. By linking light intensity with the presence or activity of tagged cells, researchers can evaluate treatment delivery, disease progression, and therapeutic responses over time.

A method for reliably grafting luciferase-tagged human malignant peripheral nerve sheath tumor cells into the sciatic nerve of immunodeficient mice is described. The use of bioluminescence imaging to demonstrate proper establishment of tumor grafts and criteria for random segregation of animals into study groups are also discussed.

Transfection into the macrophage cell line, RAW264.7, is difficult due to the cell’s natural response against foreign materials. We described here a gentle yet robust procedure for transfecting luciferase reporter genes into RAW264.7 cells.
