Lung Vasculature Flush

Lung vasculature flush is a perfusion-based procedure that removes blood and circulating material from the pulmonary vascular network, producing a cleaner lung preparation for medical and biomedical analysis. The process typically delivers a buffered physiological solution through the pulmonary circulation under controlled pressure or flow, allowing the fluid to displace intravascular blood while limiting tissue damage; the effluent is collected until the vascular bed is adequately cleared. In medicine and experimental research, this preparation improves visualization of lung structure, reduces blood-related interference in histology and molecular assays, and supports studies of pulmonary injury, inflammation, vascular remodeling, and treatment response. Consistent flushing conditions are essential for reproducible results.

We have developed a decellularized lung extracellular matrix and novel biomimetic bioreactor that can be used to generate functional lung tissue. By seeding cells into the matrix and culturing in the bioreactor, we generate tissue that demonstrates effective gas exchange when transplanted in vivo for short periods of time.

We show here the in vivo insertion of an osmotic pump for constant local drug delivery and the creation of hindlimb ischemia in a mouse model. Moreover, the hindlimb vasculature is perfused with Microfil, a silicone radiopaque agent, to prepare for micro-computed tomography (micro-CT) imaging.
