Mouse Retinal Ischemia

Mouse retinal ischemia is an experimental model in which reduced or interrupted blood flow deprives retinal tissue of oxygen and nutrients, producing injury relevant to human eye disease and ischemic brain damage. Researchers commonly induce transient ischemia by elevating intraocular pressure or obstructing retinal circulation, then examine how hypoxia causes energy failure, excitotoxic signaling, inflammation, and retinal ganglion cell loss during reperfusion. This model helps clarify the cellular mechanisms of ischemia-reperfusion injury and provides a controlled system for evaluating neuroprotective drugs, vascular therapies, and strategies to preserve vision in conditions such as retinal vascular occlusion, glaucoma, and diabetic retinopathy.

We demonstrate an in vivo electroporation protocol for transfecting single or small clusters of retinal ganglion cells (RGCs) and other retinal cell types in postnatal mice over a wide range of ages. The ability to label and genetically manipulate postnatal RGCs in vivo is a powerful tool for developmental studies.
