Optical Fractionator Method

The optical fractionator method is an unbiased stereological technique for estimating the total number of cells or other structures in a three-dimensional tissue volume, making it valuable for quantitative medical research. It combines systematic, uniform random sampling of tissue sections with optical disectors, which count structures within a defined three-dimensional probe while guard zones reduce errors from section surfaces. Because estimates rely on counting rules and sampling fractions rather than assumptions about cell size, shape, or distribution, the method supports robust comparisons across healthy and diseased tissues. Applications include assessing neuronal loss, tumor cell populations, tissue regeneration, and treatment-related cellular changes.

Here, we present a stereological method, the optical fractionator, used to quantify the formation of new neurons, and their survival, in the rat hippocampus following electroconvulsive stimulation. When correctly implemented, the sensitivity and efficiency of stereological methods ensures accurate estimates with a fixed and predetermined precision.
