Pluripotency Confirmation

Pluripotency confirmation is the assessment of whether stem cells can self-renew and differentiate into derivatives of all three embryonic germ layers, a key requirement for reliable biomedical research and therapeutic development. Researchers typically combine molecular assays for pluripotency markers such as OCT4, SOX2, and NANOG with functional tests that demonstrate differentiation into ectoderm, mesoderm, and endoderm lineages. In medicine, these evaluations help authenticate embryonic stem cells and induced pluripotent stem cells, monitor reprogramming quality, and identify cell lines suitable for disease modeling, drug screening, and regenerative applications. Rigorous confirmation also supports reproducibility and helps assess the safety of candidate cell-based therapies.

Confirmation Biases

2020

The confirmation bias is the tendency to focus on information that confirms our existing beliefs and ignore information that is inconsistent with our expectations. For example, if you think that your professor is not very nice, you notice all of the instances of rude behavior exhibited by the professor while ignoring the countless pleasant interactions he is involved in on a daily basis. Have you ever fallen prey to the confirmation bias, either as the source or target of such bias? Another...

Induced Pluripotency

Induced pluripotent stem cells (iPSCs) are somatic cells that have been genetically reprogrammed to form undifferentiated stem cells. Like embryonic stem cells, iPSCs can be grown in culture conditions that promote differentiation into different cell types. Thus, iPSCs may provide a potentially unlimited source of any human cell type, which is a major breakthrough in the field of regenerative medicine. However, more research into the derivation and differentiation of iPSCs is still needed to...

Humane Euthanasia and Confirmation of Death in Rodents

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Humane endpoints in rodents are predefined clinical, behavioral, or physiological indicators that signal an animal is experiencing significant pain, distress, or declining health. When these signs are observed, timely...

Induced Pluripotent Stem Cells

Stem cells are undifferentiated cells that divide and produce different types of cells. Ordinarily, cells that have differentiated into a specific cell type are post-mitotic—that is, they no longer divide. However, scientists have found a way to reprogram these mature cells so that they “de-differentiate” and return to an unspecialized, proliferative state. These cells are also pluripotent like embryonic stem cells—able to produce all cell types—and are therefore called induced pluripotent stem...

Source: Hiller, T. et. al. Study of Viral Vectors in a Three-dimensional Liver Model Repopulated with the Human Hepatocellular Carcinoma Cell Line HepG2. J. Vis. Exp. (2016)This video demonstrates immunohistochemical staining to evaluate viral gene transfer in a recellularized rat liver model. Reporter expression, confirmed by antibody detection and co-localization with nuclear staining, indicates successful viral transduction in the cells.
