Postoperative Analgesic Requirements

Postoperative analgesic requirements are the type and amount of pain medication a patient needs after surgery to achieve comfortable recovery while limiting adverse effects. These requirements depend on surgical tissue injury, inflammatory signaling, pain severity, individual sensitivity, and factors such as previous opioid exposure; clinicians adjust treatment through repeated pain assessments and monitoring of sedation, breathing, and function. Management may combine nonopioid medicines, regional or local anesthesia, and opioids when necessary, alongside nonpharmacological measures. Understanding postoperative analgesic requirements supports individualized prescribing, early mobilization, improved participation in rehabilitation, and safer efforts to prevent inadequate pain control, excessive sedation, and persistent opioid use.

Assessment of Morphine-induced Hyperalgesia and Analgesic Tolerance in Mice Using Thermal and Mechanical Nociceptive Modalities

We describe a protocol to examine the development of opioid-induced hyperalgesia and tolerance in mice. Based on the measurement of thermal and mechanical nociceptive responses of naïve and morphine-treated animals, it allows to quantify the increase in pain sensitivity (hyperalgesia) and decrease in analgesia (tolerance) associated with chronic opiate administration.
