Quantitative Bone Measurement

Quantitative bone measurement encompasses methods that assess bone mass, density, geometry, and strength numerically, supporting evaluation of skeletal health in medicine. Techniques such as dual-energy X-ray absorptiometry, quantitative computed tomography, and ultrasound derive measurements from X-ray attenuation, three-dimensional imaging, or sound-wave transmission, while analysis may distinguish cortical and trabecular bone. These data help clinicians identify low bone density, estimate fracture risk, monitor treatment, and assess changes associated with aging or disease. Standardized measurements also support research on osteoporosis, bone development, and responses to therapeutic interventions.

This methodology article presents a software-assisted quantitative measurement protocol to quantify histologic subchondral bone thickness in murine osteoarthritic knee joints and normal knee joints as controls. This protocol is highly sensitive to subtle thickening and is suitable for detecting early osteoarthritic subchondral bone changes.

To understand a link between the immune response and behavior, we describe a method to measure locomotor behavior in Drosophila during bacterial infection as well as the ability of flies to mount an immune response by monitoring survival, bacterial load, and real-time activity of a key regulator of innate immunity, NFκB.
