Tumor Explant Method

The tumor explant method is an ex vivo technique that maintains freshly collected tumor tissue in culture to study cancer biology outside the patient. Researchers divide specimens into small fragments and place them in a supportive culture matrix or medium, allowing nutrients and oxygen to diffuse while preserving aspects of the tumor’s three-dimensional architecture and cellular interactions. This approach can retain features of the tumor microenvironment that are often lost in isolated-cell cultures. In medicine, tumor explants support investigations of tumor growth, invasion, treatment response, and drug sensitivity, providing a clinically relevant model for translational cancer research and personalized therapy development.

Method for Novel Anti-Cancer Drug Development using Tumor Explants of Surgical Specimens

Here, we established a method for drug efficacy testing with surgical specimens of brain tumors, termed “tumor explant method”. With this method, we can evaluate drug efficacy without breaking the microenvironment of solid tumors. To validate reliability of this method, we describe representative data with our glioma specimen treated with the current first-line chemotherapeutic agent, temozolomide.
