Virus Purification

Virus purification is the process of isolating virus particles from a biological mixture to obtain a cleaner, concentrated preparation for medical and research use. It typically combines clarification to remove cells and debris with filtration, ultracentrifugation, or chromatography, which separate virions according to size, density, charge, or binding behavior while preserving infectivity or structural integrity. Purified virus preparations support vaccine manufacture, diagnostic assay development, antiviral testing, and characterization of viral structure and function. In medicine, controlling purity, concentration, sterility, and biological activity is essential for reproducible experiments, safe products, and effective quality assessment.

Source: Rghei, A. D., et al. Production of Adeno-Associated Virus Vectors in Cell Stacks for Preclinical Studies in Large Animal Models. J. Vis. Exp. (2021)The video demonstrates purification of recombinant adeno-associated virus (AAV) vectors from crude host lysate using a heparin-based affinity matrix. Viral particles bind to immobilized heparin, while impurities are washed away. A high-salt buffer releases intact viruses, which is collected in enriched fractions. These fractions are...
