White Blood Cell Analysis

White blood cell analysis is the quantitative and qualitative examination of leukocytes in blood to assess immune status and identify abnormalities. The process measures total white blood cell concentration and differentiates major cell types using automated signals such as cell size, electrical properties, light scatter, or fluorescence, often complemented by microscopic evaluation of cell morphology. In medicine, these results support the assessment of infection, inflammation, immune disorders, bone marrow dysfunction, and hematologic disease. Tracking white blood cell patterns over time can also help clinicians evaluate treatment responses and recognize changes that warrant further diagnostic testing.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.

The cell membrane modification of red blood cells (RBCs) with hyperbranched polyglycerol (HPG) is presented. Modified RBCs were characterized by aqueous two phase partitioning, osmotic fragility and complement mediated lysis. The camouflage of surface proteins and antigens was evaluated using the flow cytometry and Micro Typing System (MTS) blood phenotyping cards.
