Alpha Bungarotoxin Binding Site

An alpha-bungarotoxin binding site is the molecular region on a nicotinic acetylcholine receptor (nAChR) that recognizes alpha-bungarotoxin, a snake venom peptide used to identify receptor location and abundance in nervous tissue. The toxin binds with high affinity at the receptor’s extracellular acetylcholine-binding interface, often blocking ion-channel activation and allowing tagged toxin to mark receptors through fluorescence or other detection methods. In neuroscience, this interaction helps map nAChRs at neuromuscular junctions and synapses, distinguish receptor populations, and study cholinergic signaling, receptor organization, and changes associated with development, disease, or experimental manipulation.

Here we demonstrate the use of fluorescent Alexa dye coupled to α-bungarotoxin to measure GABA A receptor surface localization and endocytosis in hippocampal neurons. Through the use of constructs bearing a short extracellular tag that binds α-bungarotoxin, analysis of plasma membrane protein endocytic trafficking can be achieved.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.

This video demonstrates an immunofluorescence assay to detect neurodegenerative disease-specific alpha-synuclein aggregates on skin biopsy sections. Free-floating immunostaining is employed to fluorescently label the carboxyl-terminal hydrolase and the alpha-synuclein aggregates in the dermal neurons. Under a confocal microscope, cells with fluorescence from both antibodies indicate the presence of alpha-synuclein aggregates in the dermal neurons.
