Antibody Blocking

Antibody blocking is a technique that uses antibodies to bind a specific protein, receptor, or ligand and prevent its interaction or activity, helping researchers test the molecule’s role in biological systems. Blocking occurs when antibody binding sterically hinders a binding site or alters the target’s conformation, thereby reducing downstream signaling or cellular responses. In neuroscience, researchers use this approach to examine how receptors, ion channels, and signaling proteins influence neuronal communication, synaptic transmission, and behavior. Comparing blocked and unblocked conditions can reveal molecular pathways involved in neural function and disease, while also helping validate target specificity in immunoassays and functional experiments.

Chemically-blocked Antibody Microarray for Multiplexed High-throughput Profiling of Specific Protein Glycosylation in Complex Samples

In this study, we describe an improved protocol for a multiplexed high-throughput antibody microarray with lectin detection method that can be used in glycosylation profiling of specific proteins. This protocol features new reliable reagents and significantly reduces the time, cost, and lab equipment requirements as compared to the previous procedure.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

Development of a dual-functional conjugate of antigenic peptide and Fc-III mimetics (DCAF) is novel for the elimination of harmful antibodies. Here, we describe a detailed protocol for the synthesis of DCAF1 molecule, which can selectively block 4G2 antibody to eliminate antibody dependent enhancement effect during Dengue virus infection.
