Artificial Microrna

Artificial microRNAs are engineered small RNA sequences designed to reproduce the gene-silencing activity of endogenous microRNAs. In cells, an artificial microRNA is commonly expressed from a precursor transcript, processed by Drosha and Dicer, and loaded into the RNA-induced silencing complex, which uses sequence complementarity to repress target messenger RNAs through degradation or reduced translation. In neuroscience, these molecules enable targeted control of genes involved in neuronal development, synaptic signaling, and neurodegenerative disease pathways. Their programmable sequences support mechanistic studies and may improve gene-silencing strategies for experimental models and future therapies.

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns—non-coding regions of a gene—or intergenic regions—stretches of DNA present between genes. Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA ends...

Ectopic expression is one technique to elucidate the microRNAs role in brain development. However, targeting specific areas using in ovo electroporation is challenging. Here, we show an efficient way to selectively electroporate ventral and dorsal midbrain regions.

Natural Selection and Adaptive Evolution In natural settings, adaptive evolution takes place by natural selection, a process in which differences in traits lead to differences in survival and reproductive success between individuals. This happens because individuals with certain phenotypes have a higher probability of survival and/or produce more surviving offspring than individuals with other phenotypes. A classic example of this was demonstrated by Rosemary and Peter Grant studying the...

We describe here a method for regulating picornavirus tropism by incorporating sequences complementary to specific microRNAs into the viral genome. This protocol can be adapted to all different classes of viruses with modifications based upon the length and nature of their life cycle.
