Controlled Thawing

Controlled thawing is the carefully managed warming of cryopreserved biological material to restore viability while limiting damage caused by ice formation, temperature shifts, and osmotic stress. The process uses a defined warming rate, often followed by the gradual dilution and removal of cryoprotective agents, to reduce ice recrystallization and abrupt changes in cell volume. In neuroscience, controlled thawing supports the recovery of cryopreserved neurons, neural stem cells, brain organoids, and other cellular models for culture and experimentation. Consistent thawing improves post-thaw survival and reproducibility, helping researchers study neural development, disease mechanisms, cellular function, and potential regenerative therapies.

Degassing Liquids with Freeze-Pump-Thaw Cycling

Source: Laboratory of Dr. Neil Branda — Simon Fraser University Degassing refers to the process by which dissolved gases are removed from a liquid. The presence of dissolved gases such as oxygen or carbon dioxide can impede chemical reactions that utilize sensitive reagents, interfere with spectroscopic measurements, or can induce unwanted bubble formation. A number of different techniques are available for degassing liquids; some of these include heating, ultrasonic agitation, chemical...

Since James Thomson et al developed a technique in 1998 to isolate and grow hES in culture, freezing cells for later use and thawing and expanding cells from a frozen stock have become important procedures performed in routine hES cell culture. Since hES cells are very sensitive to the stresses of freezing and thawing, special care must taken. Here we demonstrate the proper technique for rapidly thawing hES cells from liquid nitrogen stocks, plating them on mouse embryonic feeder cells, and...
