Dendrite Formation

Dendrite formation is the developmental process through which neurons grow and organize branched extensions that receive signals from other cells, making it central to neural circuit assembly and function. It begins as cytoskeletal remodeling drives neurite growth, branching, and stabilization, while molecular guidance cues and neuronal activity regulate dendritic architecture and the positioning of synaptic contacts. In neuroscience, studying dendrite formation helps explain how developing brains establish connectivity and how experience shapes neural networks. It also provides insight into neurodevelopmental disorders, injury-related repair, and potential strategies for restoring disrupted communication between neurons.

Here, we show how to analyze dendritic routing of Drosophila medulla neurons in columns and layers. The workflow includes a dual-view imaging technique to improve the image quality and computational tools for tracing, registering dendritic arbors to the reference column array and for analyzing the dendritic structures in 3D space.

An imaging technique for monitoring of membrane potential changes with sub-micrometer spatial and sub-millisecond temporal resolution is described. The technique, based on laser excitation of voltage-sensitive dyes, allows measurements of signals in axons and axon collaterals, terminal dendritic branches, and individual dendritic spines.

T Cells Capture Bacteria by Transinfection from Dendritic Cells

Here a protocol is presented to measure bacterial capture by CD4+ T cells which occurs during antigen presentation via transinfection from pre-infected dendritic cells (DC). We show how to perform the necessary steps: isolation of primary cells, infection of DC, DC/T cell conjugate formation, and measurement of bacterial T cell transfection.
