Dii Injection

DiI injection is a neuroanatomical tracing method that uses the lipophilic fluorescent dye DiI to label cell membranes and map neuronal connections. After delivery into a selected region of nervous tissue, DiI inserts into lipid bilayers and spreads along interconnected membranes through lateral diffusion, allowing labeled axons and projections to be visualized by fluorescence microscopy. This technique supports the study of neural pathways, brain development, and circuit organization, particularly when researchers need to follow projections across tissue over time. DiI labeling can reveal connectivity patterns that contribute to understanding neuronal structure and functional organization.

Dual Labeling of Neural Crest Cells and Blood Vessels Within Chicken Embryos Using ChickGFP Neural Tube Grafting and Carbocyanine Dye DiI Injection

Here we report dual labeling of neural crest cells and blood vessels using chickGFP neural tube intraspecies grafting combined with intra-vascular DiI injection. This experimental technique allows us to simultaneously visualize and study development of the NCC-derived (enteric) nervous system and the vascular system, during organogenesis.

A method by which gene expression in the neural tube can be downregulated in a cell type-specific, traceable manner is described. We demonstrate how in ovo electroporation of microRNA-based plasmids that elicit spatiotemporally controlled RNA interference can be used to investigate commissural axon guidance in the developing neural tube.

Visualization of Caenorhabditis elegans Cuticular Structures Using the Lipophilic Vital Dye DiI

We present a method to visualize cuticle in live C. elegans using the red fluorescent lipophilic dye DiI (1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate), which is commonly used in C. elegans to visualize environmentally exposed neurons. With this optimized protocol, alae and annular cuticular structures are stained by DiI and observed using compound microscopy.
