Ethanol Preference Measurement

Ethanol preference measurement is a behavioral approach for quantifying an animal’s voluntary choice between ethanol-containing and water solutions, providing a readout of alcohol-related reward seeking and consumption. In a common two-bottle choice assay, animals receive simultaneous access to ethanol and water, and preference is calculated from relative fluid intake alongside total consumption and experimental conditions. In neuroscience, this measure helps link behavioral responses to genetic factors, brain circuits, and pharmacological manipulations involved in motivation and addiction. Results can reveal changes in alcohol reward sensitivity, support preclinical evaluation of candidate treatments, and improve interpretation of substance use disorder models.

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

C. elegans is a useful model for studying the effects of ethanol on behavior. We present a behavioral assay that quantifies the effects of ethanol on the locomotion speed of crawling worms; both initial sensitivity and the development of acute functional tolerance to ethanol can be measured with this assay.
