Ex Vivo Neuronal Preparation

Ex vivo neuronal preparation is a neuroscience method in which neurons or neural tissue are removed from an organism and maintained under controlled laboratory conditions for direct study. Researchers typically preserve viable tissue in oxygenated physiological solutions that supply essential ions and nutrients, allowing neuronal circuits to retain electrical activity and synaptic function outside the body. These preparations support electrophysiological recording, calcium or fluorescence imaging, pharmacological manipulation, and analysis of synaptic transmission. By isolating defined neural circuits while providing experimental control, ex vivo preparations help reveal how neurons communicate, how drugs alter network activity, and how cellular mechanisms contribute to behavior and disease.

With its small transparent body, well-documented neuroanatomy and a host of amenable genetic techniques and reagents, C. elegans makes an ideal model organism for in vivo neuronal imaging using relatively simple, low-cost techniques. Here we describe single neuron imaging within intact adult animals using genetically encoded fluorescent calcium indicators.

This video shows a procedure for generating neuronal cultures from late embryo and early postnatal mouse cortex.
