Functional Synaptic Mapping

Functional synaptic mapping is the systematic identification and characterization of neuronal connections by measuring how activity in one cell or circuit influences another. It commonly combines targeted stimulation, such as activating defined axons, with electrophysiological recording of postsynaptic responses; response timing, amplitude, and pharmacological sensitivity help distinguish direct synaptic inputs and assess their excitatory or inhibitory effects. In neuroscience, these maps reveal circuit organization, connection strength, and cell-type-specific communication. They support studies of sensory processing, learning, development, and neurological disease while helping researchers link synaptic architecture to network function and behavior.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

Here, we describe a standardized protocol for motor mapping using nTMS combined with diffusion tensor imaging (DTI)-based reconstruction of the corticospinal tract (CST). The protocol is reproducible, clinically feasible, and readily integrable into routine clinical workflows, providing a robust and valuable framework for motor pathway assessment, neuroplasticity research, and rehabilitation planning.
