Honeybee Brain Recording

Honeybee brain recording is a neuroscience technique for measuring neural activity in Apis mellifera, linking brain function to sensory processing and behavior. Researchers place fine electrodes near selected neurons or brain regions and record changes in electrical signals while bees receive stimuli or perform tasks. These recordings help reveal how honeybees process odors, colors, movement, and social cues, and how neural circuits support learning, memory, navigation, and decision-making. Because honeybees combine complex behavior with a compact, experimentally accessible nervous system, brain recordings provide valuable models for studying neural coding, behavioral adaptation, and the biological basis of cognition.

Simultaneous extracellular long term recordings from two different brain neuropiles or two different anatomical tracts were established in honeybees. These recordings allow the investigation of temporal aspects of neuronal processing across different brain areas at the single neuron as well as at the ensemble level in a behaving animal.

To test the inhibitory effects of pharmacologic agents on phospholipase C (PLC) in different regions of the honeybee brain, we present a biochemical assay to measure PLC activity in those regions. This assay could be useful for comparing PLC activity among tissues, as well as among bees exhibiting different behaviors.

Here we describe our detailed protocol for the preparation of single-cohort honeybee colonies – a useful tool for analyzing the role-associated worker physiology. We also describe detailed protocols for treating workers with juvenile hormone and ecdysone to evaluate the involvement of these hormones in the regulation of worker behavior and/or physiology.

We demonstrate how to implement a behavioral pharmacology method in an appetitive olfactory conditioning paradigm in honeybees (Apis mellifera) by systemic application of drugs. This method allows investigation of the mechanisms underlying learning and memory formation in a simple and reliable way.
