Live Cell Staining

Live cell staining is a method for labeling cells with dyes or fluorescent probes while they remain viable, enabling researchers to visualize cellular structures, functions, and behavior in real time. Stains cross the plasma membrane or enter through transport pathways, then bind specific molecules, accumulate in organelles, or undergo enzyme-dependent conversion to produce detectable fluorescence or color without immediately disrupting cell physiology. In neuroscience, live cell staining helps reveal neuronal morphology, membrane integrity, organelle activity, and dynamic processes such as calcium signaling or axonal transport. These observations support studies of neural development, cell injury, disease mechanisms, and responses to experimental treatments.

We describe a streamlined protocol for generating "fillet" preparations of Drosophila embryos of specific genotypes. This protocol allows efficient execution of a variety of genetic screens. It also allows excellent visualization of structures in the late embryo.
