Outer Hair Cells

Outer hair cells are specialized sensory cells in the cochlea that amplify sound-induced vibrations and sharpen hearing sensitivity, making them central to auditory neuroscience. When movement of the basilar membrane bends their stereocilia, mechanotransduction channels alter the cells’ membrane potential; the motor protein prestin then drives rapid changes in cell length that feed mechanical energy back into the cochlear partition. This cochlear amplification improves the detection of quiet sounds and supports precise frequency selectivity. Because outer hair cells are vulnerable to noise, aging, and ototoxic drugs, their structure and function provide important models for studying sensorineural hearing loss and developing protective or restorative therapies.

2011

A reliable method to investigate outer hair cell (OHC) motile responses, including electromotility, slow motility and bending, is described. OHC motility is elicited by stimulation with an external alternating electrical field, and the method takes advantage of high-speed image recording, LED-based illumination, and last generation image analysis software.

Hair cells are the sensory receptors of the auditory system—they transduce mechanical sound waves into electrical energy that the nervous system can understand. Hair cells are located in the organ of Corti within the cochlea of the inner ear, between the basilar and tectorial membranes. The actual sensory receptors are called inner hair cells. The outer hair cells serve other functions, such as sound amplification in the cochlea, and are not discussed in detail here. Hair cells are named after...

Source: Lukasz, D., et al. In Vivo Calcium Imaging of Lateral-line Hair Cells in Larval Zebrafish. J. Vis. Exp. (2018)This video demonstrates live imaging of calcium influx in the lateral-line hair cells of a paralyzed transgenic zebrafish larva using a confocal microscope. Mechanical stimulation of hair bundles via fluid pulses induces calcium entry through mechanotransduction channels and voltage-gated channels, which is visualized in real-time using fluorescence indicators. The method...

Modified Experimental Conditions for Noise-Induced Hearing Loss in Mice and Assessment of Hearing Function and Outer Hair Cell Damage

2023

Here, we present a protocol for a mouse model of noise-induced hearing loss (NIHL). To induce NIHL, we developed a new and simple device using corrugated plastic, a rat trap cage, and a speaker. Auditory brainstem response and immunofluorescence imaging were employed to assess the hearing function and outer hair cell damage, respectively.
