Peroxidase-tagged Antibodies

Peroxidase-tagged antibodies are immunological detection reagents that combine antigen-specific antibodies with an enzyme, most commonly horseradish peroxidase, to reveal target molecules in cells or tissue. After the antibody binds its antigen, the peroxidase catalyzes oxidation of a supplied substrate, producing a colored, insoluble precipitate or a chemiluminescent signal that can be detected by microscopy or imaging systems. In neuroscience, these reagents support immunohistochemistry and related assays for mapping proteins, neurotransmitter-associated markers, and cellular structures in nervous tissue. Their high specificity and signal amplification enable sensitive localization studies in research on neural organization, development, and disease.

Fast and Specific Assessment of the Halogenating Peroxidase Activity in Leukocyte-enriched Blood Samples

This protocol describes the quick enrichment of leukocytes from small blood samples for a subsequent specific determination of the halogenating peroxidase activity within the cells. The method can be applied to human and non-human material and may contribute to the evaluation of new inflammatory markers.

A Multiplex Immunostaining Method Using Primary Antibodies from the Same Host Species

This video demonstrates an assay to perform multiplex immunostaining using primary antibodies from the same host species. In this method, an adrenal tissue section undergoes a two-step immunostaining process. Initially, the section is stained with antibodies specific for markers in the cortex, followed by stripping the bound antibody and restaining with antibodies specific for markers in the medulla.

This paper presents an economical and efficient protocol for examining peroxidase-positive leukocytes in semen. With the assistance of a computer-assisted semen analysis (CASA) system, the concentration of peroxidase-positive leukocytes in semen can be obtained within a total of 60 min, effectively improving the efficiency of andrology laboratory and andrologists.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
