Primary Motoneuron Enrichment

Primary motoneuron enrichment is a cell-isolation technique that increases the proportion of motoneurons in cultures derived from neural tissue, providing a more defined model for neuroscience research. The process typically involves dissociating spinal cord tissue and separating motoneurons from other cell types through selective adhesion, immuno-based capture, or differential survival under controlled culture conditions. Enriched cultures support investigations of neuronal development, axon growth, synaptic function, and vulnerability to injury or neurotoxic compounds. They are especially valuable for studying motoneuron diseases, including amyotrophic lateral sclerosis, and for evaluating potential neuroprotective or regenerative therapies in experimentally manageable systems.

Lectin-based Isolation and Culture of Mouse Embryonic Motoneurons

An alternative way of isolating mouse embryonic motoneurons from the spinal cord is described. The method takes into account the fact that lectin can bind to the low affinity nerve growth factor receptor p75NTR. This lectin-based preplating allows a purification similar to that with a specific antibody against the p75NTR.
