Pro-inflammatory Model

A pro-inflammatory model is an experimental system designed to reproduce or study immune activation and inflammatory responses under controlled conditions. In neuroscience, these models typically stimulate inflammatory signaling in neural or supporting cells, leading to the release of cytokines and other mediators that can alter neuronal function, glial activity, and tissue homeostasis. Researchers use pro-inflammatory models to investigate how inflammation contributes to neurodegeneration, pain, infection, injury, and neurological disease, while evaluating potential therapeutic strategies. By linking molecular signals with cellular and behavioral outcomes, these models help clarify disease mechanisms and support the development and testing of anti-inflammatory interventions.

DNBS/TNBS induced colitis offers an alternative and inexpensive method to study the pathobiology of inflammatory bowel disease. The protocol discussed in this paper outlines the successful application of DNBS to induce colitis in mice and rats and allows one to thoroughly study host-mediated intestinal responses.

Generating a Pro-Inflammatory Organ Culture Model to Simulate an Early Intervertebral Disc Disease

2025

This video demonstrates a technique for culturing bovine intervertebral discs in bioreactors to develop a three-dimensional organ culture model. Specific nutrient-deprived media and degenerative pressure conditions are applied to the discs, mimicking the pro-inflammatory and degenerative microenvironment observed during early-stage intervertebral disc disease. The disc heights are measured post-treatment to analyze the effects of nutrient deprivation and degenerative loading.

Microfluidic flow chambers etched by photolithography and fabricated from PDMS are applied to probe functional outcomes associated with EC dysfunction and inflammation. In a representative experiment, the ability of differential shear stress to modulate monocytic cell adhesion to cytokine activated EC monolayers is demonstrated.
