Prp Mrna Cleavage

PrP mRNA cleavage is the regulated cutting of messenger RNA that encodes the cellular prion protein, a process relevant to post-transcriptional gene control in the nervous system. Specific RNA-cleaving enzymes or regulatory complexes recognize sequence or structural features in the transcript, producing fragments that can alter mRNA stability, translation, or subsequent degradation. Studying this process helps clarify how neurons control PrP expression and how disruptions in RNA regulation may influence prion biology and neurodegenerative disease. Experimental analysis of cleavage sites and resulting RNA products can therefore support research into neuronal gene regulation, disease mechanisms, and potential molecular interventions.

Implant-associated infection is a significant clinical complication. This study describes an approach using platelet-rich plasma (PRP) to prevent implant-associated infections, presents the protocol for preparing PRP with constant platelet concentration, and reports the newly identified antimicrobial properties of PRP and related protocols for examining such antimicrobial properties in vitro.

RNA polymerase II synthesizes a precursor RNA that extends beyond the 3' end of the mature mRNA. The end of the mature RNA is generated cotranscriptionally, at a site dictated by RNA sequences, via the endonuclease activity of the cleavage complex. Here, we detail the method to study cleavage reactions in vitro.
