Rpe Isolation

RPE isolation is a laboratory technique used to separate retinal pigment epithelial cells from surrounding ocular tissues for structural, molecular, and functional studies. The process typically combines careful dissection of the retina with enzymatic or mechanical separation, preserving the RPE sheet or generating individual cells for culture and analysis. In neuroscience and vision research, isolated RPE preparations help investigators examine the blood-retina barrier, photoreceptor support, retinal metabolism, inflammation, and responses to disease-related stress. The method supports gene-expression studies, imaging, drug testing, and investigations of retinal disorders, providing a controlled system for linking RPE dysfunction to neuronal degeneration.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
