Serial Tissue Sections

Serial tissue sections are consecutive thin slices collected in sequence to preserve the spatial organization of a specimen, making them valuable for mapping structures that extend across multiple planes. In neuroscience, tissue is embedded, sectioned at a consistent thickness, and carefully ordered so that stained or labeled features can be compared from one section to the next and reconstructed in three dimensions. This approach supports analysis of brain architecture, neuronal distributions, lesion boundaries, and connections between regions. By linking microscopic observations to their position within the intact tissue, serial sections provide a foundation for anatomical reconstruction and quantitative studies of nervous-system organization.

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.
