Small-scale Fractionation

Small-scale fractionation is a laboratory method for separating a small amount of biological material into subcellular or biochemical fractions, making it useful for analyzing complex tissues such as the brain. In neuroscience, tissue is gently homogenized to release cellular components, then subjected to sequential centrifugation steps that separate nuclei, membranes, synaptic material, and soluble components according to size and density. The resulting fractions can be examined using biochemical, molecular, or imaging assays to determine where proteins, receptors, enzymes, or signaling molecules are located. This approach supports studies of synaptic organization, neuronal signaling, disease-associated changes, and cellular responses when sample availability is limited.

Ribosomes play a central role in protein synthesis. Polyribosome (polysome) fractionation by sucrose density gradient centrifugation allows direct determination of translation efficiencies of individual mRNAs on a genome-wide scale. In addition, this method can be used for biochemical analysis of ribosome- and polysome-associated factors such as chaperones and signaling molecules.
