Surgical Stroke Induction

Surgical stroke induction is an experimental method that creates a controlled cerebral ischemic or hemorrhagic injury through a surgical procedure, enabling researchers to study stroke under defined conditions. Depending on the model, surgery can disrupt a cerebral blood vessel or alter local brain tissue, reducing blood flow or causing bleeding and triggering neuronal injury, inflammation, and subsequent tissue damage. In neuroscience, these models support investigations of stroke mechanisms, brain repair, behavioral outcomes, and therapeutic strategies. By standardizing injury location, severity, and timing, surgical stroke induction helps researchers compare disease processes and evaluate potential interventions before clinical studies.

In order to understand the pathophysiology of stroke, it is important to use reliable models. This paper will describe one of the most frequently used stroke models in mice, termed the middle cerebral artery occlusion (MCAo) model (also termed the intraluminal filament or suture model) with reperfusion.

University of Notre Dame, IN The Guide for the Care and Use of Laboratory Animals ("The Guide") states that pain assessment and alleviation are integral components of the veterinary care of laboratory animals. The definition of anesthesia is the loss of feeling or sensation. It is a dynamic event involving changes in anesthetic depth with respect to an animal's metabolism, surgical stimulation, or variations in the...

Various murine models of middle cerebral artery occlusion (MCAO) are widely used in experimental brain research. Here, we demonstrate the model of transcranial permanent distal MCAO which produces consistent cortical infarction of a size corresponding to damage imposed by the majority of human ischemic strokes.

Filament Insertion-based Transient Stroke Model: A Surgical Procedure to Perform Middle Cerebral Artery Occlusion in Murine Brain

2025

This video describes the procedure to develop transient stroke in a murine model using silicon-coated filament-based occlusion of the middle cerebral artery. The developed model mimics human stroke, thus helping to investigate novel therapeutic strategies for treating stroke.
