Hyperlipidemia Treatment

Hyperlipidemia treatment comprises lifestyle measures and medications that reduce abnormally elevated blood lipids, especially low-density lipoprotein cholesterol (LDL-C), to lower the risk of atherosclerotic cardiovascular disease. Pharmacologic therapy commonly targets hepatic cholesterol handling: statins inhibit HMG-CoA reductase, reducing cholesterol synthesis and increasing hepatic LDL-receptor expression, while ezetimibe limits intestinal cholesterol absorption and PCSK9 inhibitors enhance LDL-receptor recycling. Treatment selection depends on lipid levels, cardiovascular risk, comorbidities, and response to therapy. Effective management can reduce circulating atherogenic lipoproteins, slow plaque development, and prevent myocardial infarction and stroke.

This protocol presents a detailed approach to replicating a murine model of hyperlipidemia-induced heart failure with preserved ejection fraction (HFpEF). The design combines the administration of adeno-associated virus 9-cardiac troponin T-low-density lipoprotein receptor (AAV9-cTnT-LDLR) and poloxamer-407 (P-407).
