Tissue Plasminogen Activator

Tissue plasminogen activator (tPA) is a serine protease that regulates fibrinolysis, the physiological breakdown of blood clots, and serves as an important pharmacological therapy. It binds fibrin within a thrombus and catalyzes the conversion of plasminogen to plasmin, which degrades fibrin strands and can restore blood flow. Recombinant forms such as alteplase and modified agents such as tenecteplase are administered for selected acute thrombotic conditions, including ischemic stroke and myocardial infarction, under strict timing and safety criteria. Pharmacological study of tPA addresses dosing, delivery, clot selectivity, and bleeding risk, supporting safer thrombolytic treatment and next-generation fibrinolytic drugs.
