Ldl Isolation

LDL isolation is a biochemical method for separating low-density lipoprotein particles from plasma or serum so their composition, structure, and biological activity can be studied. In density-gradient ultracentrifugation, samples are adjusted with a density medium and spun at high speed, allowing LDL to form a distinct fraction according to its buoyant density, which can then be collected and analyzed. Isolated LDL supports research on lipid transport, cholesterol metabolism, lipoprotein-receptor interactions, and oxidative modification. The method also provides controlled material for investigating cardiovascular disease mechanisms and evaluating biochemical assays or therapeutic strategies.

We have developed a simplified method to rapidly isolate purified HDL suitable for miRNA analysis from human plasma.

This protocol provides an efficient approach to measuring LDL cholesterol uptake with real time influx rates using a live cell imaging system in various cell types. This technique provides a platform to screen the pharmacological activity of compounds affecting LDL influx while monitoring for cell morphology and hence potential cytotoxicity.

The vascular endothelial cells play a significant role in many important cardiovascular disorders. This article describes a simple method to isolate and expand endothelial cells from the mouse aorta without using any special equipment. Our protocol provides an effective means of identifying mechanisms in endothelial cell physiopathology.
