Phospholipase Assay

A phospholipase assay is a biochemical method used to measure enzymes that hydrolyze phospholipids, revealing how lipid substrates are converted into signaling molecules and other products. In a typical assay, a phospholipase acts on a defined phospholipid substrate, cleaving specific ester or phosphodiester bonds; the reaction is quantified by detecting released fatty acids, phosphate-containing products, or changes in a linked colorimetric, fluorescent, or radiometric signal. These assays help distinguish phospholipase activities, characterize enzyme kinetics and substrate specificity, and evaluate effects of inhibitors, mutations, pH, cofactors, or detergents. They support research in membrane biology, lipid signaling, pharmacology, and disease mechanisms.

In this video, we demonstrate an assay to establish the presence of phospholipase in the crude venom extract from Anthopleura dowii—a marine sea anemone. The assay utilizes an agarose gel diffusion technique, in which the enzyme phospholipase diffuses and breaks down phospholipids incorporated in the solid media in a radial manner.
