Endpoint Format Analysis

Endpoint Format Analysis is an analytical approach that evaluates a biological or biochemical system by measuring its final response after a defined experimental period. In this format, samples undergo controlled assay conditions until a specified endpoint, after which a measurable signal, such as absorbance, fluorescence, or another assay output, is compared with controls or reference standards. In bioengineering, endpoint measurements support the evaluation of enzyme activity, biomolecular interactions, cell responses, and engineered biological systems. The approach can simplify data collection, enable comparisons across conditions, and help assess assay performance, although it provides less information about reaction dynamics than continuous or kinetic analysis.

We describe a method for the qualitative and quantitative analysis of stress granule formation in mammalian cells after the cells are challenged with bacteria and a number of different stresses. This protocol can be applied to investigate the cellular stress granule response in a wide range of host-bacterial interactions.

We provide a simple, semi-quantitative method to investigate biofilm formation in vitro. This method takes advantage of the Zeiss stemi 2000-C Dissecting Microscope (with camera attachment) to monitor both the timing and pattern of biofilm formation, as assessed by the development of wrinkled colonies.
