Fibrotic Disease Models

Fibrotic disease models are experimental systems that reproduce the excessive deposition and remodeling of extracellular matrix characteristic of fibrosis, enabling researchers to study how healthy tissue becomes stiff, scarred, and dysfunctional. In bioengineering, these models combine cells, biomaterials, and controlled biochemical or mechanical cues to activate pathways such as transforming growth factor beta signaling, promote fibroblast-to-myofibroblast transition, and generate collagen-rich tissue environments. Two-dimensional cultures, three-dimensional matrices, organoids, and engineered tissues can model fibrosis in organs including the lung, liver, heart, and kidney. They support investigation of disease mechanisms, evaluation of antifibrotic therapies, and development of more physiologically relevant platforms for precision medicine.

Sustained fibrosis with deposition of excessive extracellular matrix proteins leads to cirrhosis. Alcohol abuse is one of the main causes of severe liver disease. We established an ethanol-induced zebrafish fibrotic liver model to study the mechanisms and strategies of promoting hepatocyte regeneration upon alcohol-induced injury.

Mouse model of metabolic dysfunction-associated steatotic liver disease (MASLD) with metabolic dysfunction, hepatic gene expression changes, and liver histopathological alterations that resemble human MASLD, including fibrosis that progresses to advanced fibrosis stage 3. This model can be used in studies of MASLD pathophysiology and in pre-clinical studies of new therapies.

The inoculation of Trypanosoma cruzi in fertile eggs prior to incubation renders the parasite kDNA minicircle integration in embryo cells genome. Crossbreeding reveals the vertical transfer of the mutations to progeny. The kDNA integrates into coding regions at several chromosomes and the chickens die with an inflammatory autoimmune heart disease.

This video demonstrated a protocol for intravitreous injection using a 1ml syringe with glass pipette. Useful tips about avoiding massive bleeding and lens damage are given.
