Localized Therapeutics

Localized therapeutics are treatments engineered to deliver a drug or biologic directly to a diseased tissue or anatomical site, concentrating therapeutic activity where it is needed while reducing exposure elsewhere. In bioengineering, this approach uses vehicles or devices such as injectable hydrogels, drug-eluting implants, nanoparticles, or engineered scaffolds that retain, release, or transport therapeutic agents through processes including degradation, diffusion, or responses to local conditions. Localized delivery can improve efficacy, reduce systemic toxicity, and support tissue repair, making it valuable for cancer therapy, regenerative medicine, infection control, and precision treatment. Research continues to refine dose, timing, spatial distribution, and compatibility with host tissue.

Bioimaging methods used to assess cell biodistribution of nanoparticles are applicable for therapeutic and diagnostic monitoring of nanoformulated compounds. The methods described herein are sensitive and specific when assessed by histological coregistration. The methodologies provide a translational pathway from rodent to human applications.

We describe the method of programming stem cells to overexpress therapeutic factors for angiogenesis using biodegradable polymeric nanoparticles. Processes described include polymer synthesis, transfecting adipose-derived stem cells in vitro, and validating the efficacy of programmed stem cells to promote angiogenesis in a murine hindlimb ischemia model.
