Calcium Ionophore Stimulation

Calcium ionophore stimulation is a biological technique that artificially elevates intracellular calcium levels to investigate calcium-dependent cellular functions. Calcium ionophores are lipid-soluble molecules that transport Ca2+ across cell membranes, disrupting normal calcium gradients and activating downstream signaling pathways. Researchers use this approach to examine processes such as secretion, contraction, enzyme activity, and cellular activation under controlled experimental conditions. In reproductive biology, immunology, and cell signaling studies, ionophore treatment can help reveal how calcium regulates cell behavior, although exposure time and concentration must be carefully controlled to limit toxicity or irreversible cellular changes.

A protocol for the induction of eryptosis, programmed cell death in erythrocytes, using the calcium ionophore, ionomycin, is provided. Successful eryptosis is evaluated by monitoring the localization phosphatidylserine in the membrane outer leaflet. Factors affecting the success of the protocol have been examined and optimal conditions provided.
