Coverslip Preparation

Coverslip preparation is the process of cleaning, positioning, and mounting a thin glass coverslip over a biological specimen for microscopic examination. It typically involves placing the sample on a slide, adding an aqueous or resin-based mounting medium, lowering the coverslip at an angle to reduce trapped air, and allowing the mount to set when needed. Proper preparation preserves specimen structure, improves optical clarity, and helps maintain a consistent distance between the objective lens and sample. This technique supports bright-field, fluorescence, and other microscopy applications, enabling reliable imaging of cells, tissues, microorganisms, and stained preparations.

Preparation of a Micropatterned Substrate to Study Schwann Cell Phenotypes

2025

The video describes the preparation of a micropatterned substrate to study cell phenotype specification. A protein-coated micropatterned PDMS stamp is used to transfer the micropattern on a PDMS-coated coverslip. The coverslip is then treated with a surfactant to hinder cell adhesion at the unwanted surfaces. Cells are then added to settle in the pattern and incubated for further analysis.
