Bacteriocin Production

Bacteriocin production is the biological process by which bacteria synthesize antimicrobial peptides that inhibit or kill closely related or competing microorganisms. Genes encoding bacteriocins are expressed in response to cellular and environmental signals, and the resulting peptides are often ribosomally synthesized, processed, and exported with protective mechanisms that prevent harm to the producing cell. In biology, studying this process clarifies microbial competition, communication, and community structure. Bacteriocins also support research into food preservation, probiotic development, and alternatives to conventional antibiotics, while their genetic pathways provide targets for engineering strains with defined antimicrobial properties.

Competition in Streptococcus pneumoniae is mediated by bacteriocins, small antimicrobial peptides with inhibitory activity towards pneumococcus and other related species. Here we describe an optimized bacterial overlay assay that allows for the characterization of bacteriocin activity and inhibitory spectrum, bacteriocin-specific immunity, and detection of secreted quorum sensing peptides.
