Phospholipid Films

Phospholipid films are thin layers of amphipathic molecules that organize at air-water or other interfaces, providing a model for understanding biological membranes. Their hydrophilic heads interact with water while hydrophobic tails orient away from it, driving spontaneous formation of monolayers, bilayers, or other ordered structures. In biology, researchers use phospholipid films to examine membrane organization, surface tension, molecular interactions, and the behavior of membrane-associated proteins. These systems also support studies of drug delivery, biosensors, artificial membranes, and the physical principles underlying cell boundaries and compartmentalization.

Quantitative Flow Cytometry to Study Labeled Protein-Phospholipid Vesicle Interactions

In this video, we demonstrate the interaction between proteins and artificial phospholipid vesicles using quantitative flow cytometry. The binding of fluorescently-labeled proteins to fluorescently-labeled phospholipid vesicles increases the mean fluorescence intensity, and this increase is detected by a flow cytometer.

Extraction and Analysis of Microbial Phospholipid Fatty Acids in Soils

Phospholipid fatty acids provide information about the structure of soil microbial communities. We present methods for extraction from soil samples with a single-phase chloroform mixture, fractionation of extracted lipids using solid phase extraction columns, and methanolysis to produce fatty acid methyl esters, which are analyzed by capillary gas chromatography.
