Droplet Digital Pcr

Droplet digital PCR (ddPCR) is a highly sensitive nucleic acid quantification method that measures rare DNA or RNA targets with greater precision than many conventional amplification assays. The sample is divided into thousands of nanoliter-sized droplets, each undergoing PCR independently; endpoint fluorescence identifies positive and negative droplets, while Poisson statistics convert their distribution into an absolute target concentration. In cancer research, ddPCR can detect low-frequency mutations, quantify copy-number changes, and measure circulating tumor DNA in blood samples. These capabilities support tumor profiling, treatment-response monitoring, minimal residual disease assessment, and the development of more sensitive molecular diagnostics.

Here we present a protocol to illustrate the isolation of rare cancer cells from a contrived sample containing whole blood and spiked in lung cancer cells. The protocol utilizes a semi-automated, label-free cell isolation platform. Enriched cells are subsequently analyzed for epidermal growth factor receptor mutations using droplet digital PCR.
