Single-element Resolution

Single-element resolution is the ability of an analytical method to detect and distinguish one chemical element from surrounding elements, enabling precise measurement of elemental composition. In cancer research, element-specific signals are separated by their characteristic energy, wavelength, or mass-to-charge response, allowing researchers to map where metals and other elements occur within cells, tissues, or tumors. This capability supports studies of metal homeostasis, tumor microenvironments, therapeutic compounds, and contrast agents. By linking elemental distributions with cellular structure and disease processes, single-element resolution can clarify mechanisms of cancer biology and improve the evaluation of diagnostic and treatment strategies.

We describe a protocol to volumetrically image fluorescent protein labeled cells deep inside intact embryonic and postnatal hearts. Utilizing tissue-clearing methods in combination with whole mount staining, single fluorescent protein-labeled cells inside an embryonic or postnatal heart can be imaged clearly and accurately.
