Drug Response Variability

Drug response variability is the range of differences in how individuals benefit from, tolerate, or metabolize the same medication, making it central to safe and effective clinical care. It reflects interactions among pharmacokinetics, which governs absorption, distribution, metabolism, and excretion, and pharmacodynamics, which describes drug effects at target sites; genetic variation, age, organ function, disease state, and drug interactions can alter these processes. Understanding drug response variability supports individualized dosing, selection of therapies, monitoring for adverse effects, and interpretation of treatment outcomes in clinical research and practice.

Factors Affecting Drug Response: Overview

2023

When it comes to infants and young children, they are typically administered smaller doses of medication in comparison to adults. This is primarily because their organ functions still need to fully develop, meaning their bodies are not as efficient at metabolizing or eliminating drugs. Additionally, their blood-brain barrier is more permeable than in adults. As a result, high concentrations of drugs can easily penetrate the central nervous system (CNS), potentially leading to neurological...

2016

We present a facile method to fabricate a biodegradable gelatin-based drug release platform that is magneto-thermally responsive. This was achieved by incorporating superparamagnetic iron oxide nanoparticles and poly(N-isopropylacrylamide-co-acrylamide) within a spherical gelatin micro-network crosslinked by genipin, in conjunction with an alternating magnetic field application system.

Synergistic Effect of Intracranial and Intraperitoneal Delivery of Analgesic Drugs on Thermal Pain Response in a Rat

Source: Martinez, E., et al. Intracranial Pharmacotherapy and Pain Assays in Rodents. J. Vis. Exp. (2019).This video demonstrates the intracranial administration of an analgesic drug in an anesthetized rat with a pre-implanted guide cannula in the prefrontal cortex. An additional analgesic drug is co-administered intraperitoneally. Following recovery, the rat’s thermal pain response is assessed using Hargreave’s apparatus, where paw withdrawal latency is measured to evaluate the analgesic...

Chronopharmacokinetics: Circadian Rhythms and Influence on Drug Response

2025

Circadian rhythms are cyclic changes that are crucial in plasma drug concentrations. Various standard circadian parameters, including core body temperature, heart rate, and other cardiovascular factors, directly impact disease states and the therapeutic response to drug therapy. The time of drug administration is an important factor to consider, as it can influence the toxic dose of a drug. For example, a study conducted by Prins et al. in 1997 examined the effects of the timing of...
