Endocardial Cells

Endocardial cells are specialized endothelial cells that line the heart chambers and valves, where they regulate communication between the bloodstream and developing cardiac tissues. During embryonic heart development, they respond to signals such as vascular endothelial growth factor, bone morphogenetic proteins, and Notch, and can undergo endothelial-to-mesenchymal transition to form cardiac valves and septa. Their interactions with myocardial cells also influence trabeculation, chamber formation, and cardiac maturation. Studying endocardial cells helps explain congenital heart defects and provides models for investigating cardiovascular development, disease mechanisms, tissue engineering, and potential regenerative therapies.

Classically, the endocardium of the mouse embryonic valve primordium has been analyzed using transversal, coronal, or sagittal sections. Our novel approach for en face, two-dimensional imaging of the endocardium at valvulogenic regions allows planar polarity and cell rearrangement analysis of the endocardium during valve development.
