Neutralization Medium Wash

Neutralization Medium Wash is a cell-culture procedure that removes or inactivates residual dissociation reagent after cells have been detached, helping preserve cell viability and physiological behavior. The wash uses a compatible culture medium to dilute the reagent and, when appropriate, neutralize its activity through inhibitory components, limiting continued enzymatic or chemical exposure during cell handling. In developmental biology, this step supports the recovery and maintenance of embryonic, progenitor, and differentiated cells before replating or further experimentation. Consistent washing improves reproducibility in cell expansion, differentiation assays, tissue modeling, and other studies of cellular development.

Optimization of a Quantitative Micro-neutralization Assay

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples.
