Pax6 Otx2

Pax6 and Otx2 are transcription factors that regulate gene expression during embryonic development, making them important determinants of tissue identity and organ formation. By binding regulatory DNA sequences and interacting with other developmental regulators, they activate or repress target genes in spatially and temporally controlled patterns. Pax6 is strongly associated with eye and nervous system development, whereas Otx2 helps establish anterior neural and head structures. Studying their expression, regulatory networks, and functional interactions helps explain how cells acquire specialized identities and how developmental abnormalities arise, while supporting research in neurodevelopment, congenital disease, stem cell differentiation, and tissue engineering.

Homeobox genes are regulatory genes often associated with tumors in the adult organisms. We investigated their comparative expression by immunohistochemical and real-time PCR analysis, in normal and inflammatory nasal mucosae and in sinonasal neoplasms in order to use them as possible diagnostic and therapeutic targets.

This protocol describes an efficient method of differentiating hiPSCs into eye field clusters and generating neuro-retinal organoids using simplified culture conditions involving both adherent and suspension culture systems. Other ocular cell types, such as the RPE and corneal epithelium, can also be isolated from mature eye fields in retinal cultures.
