Transgenic Reporter Mice

Transgenic reporter mice are genetically engineered animals that carry a reporter gene, enabling researchers to visualize or measure the activity of selected genes in living tissues. Typically, regulatory DNA from a gene of interest controls a fluorescent protein or enzyme such as luciferase, linking reporter output to gene expression in specific cells, developmental stages, or physiological conditions. These models help map tissue-specific expression, follow cell lineages, monitor signaling pathways, and assess responses to disease or treatment. By connecting genetic regulation with observable signals, transgenic reporter mice provide a powerful approach for studying gene function and dynamic biological processes in vivo.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

In this article we describe the principles for designing and performing a multi-color lineage tracing experiment using R26R-Confetti mice. We provide a specific protocol for tracking corneal epithelial cells which can be modified for other tissues of interest.
