3d Culture Model

A 3D culture model is an in vitro system that grows cells within a three-dimensional architecture, more closely reproducing tissue organization than flat, two-dimensional cultures. Cells interact with neighboring cells and extracellular matrix, creating spatial organization and gradients of oxygen, nutrients, signaling molecules, and infection-related factors that influence cell behavior and immune responses. In immunology and infection research, these models can represent barriers, tissues, or organ-like structures for studying pathogen entry, replication, inflammation, and host-pathogen interactions. They also support evaluation of antimicrobial or immunomodulatory treatments under conditions that may better predict tissue-level outcomes than conventional culture.

The video describes the laser dissection of the 3D organotypic co-culture model with micron-level precision. In 3D organotypic models, stromal constituents such as fibroblasts are 3-dimensionally co-cultured with cancer epithelial cells. The 3D model serves as an experimental tool that enables invasion and cancer-stroma interactions to be studied in near-physiological conditions.
