Automated Staining

Automated staining is a laboratory technique that uses instruments to apply stains or labeling reagents to biological specimens with controlled timing, volumes, and processing steps. In immunology and infection research, automated systems can dispense primary and secondary antibodies, perform timed incubations and washes, and develop colorimetric or fluorescent signals that reveal specific cells, proteins, or microbial components. By standardizing sample preparation and reducing manual handling, the method improves reproducibility, throughput, and workflow efficiency across tissue sections, cytology samples, and other specimens. These consistent results support pathogen detection, immune-cell characterization, disease investigation, and comparative analysis in research and diagnostic settings.

An Automated Differential Nuclear Staining Assay for Accurate Determination of Mitocan Cytotoxicity

The protocol describes a rapid, high-throughput, reliable, inexpensive, and unbiased assay for efficiently determining cellular viability. This assay is particularly useful when cells' mitochondria have been damaged, which interferes with other assays. The assay uses automated counting of cells stained with two nuclear dyes – Hoechst 33342 and propidium iodide.

Peptide adsorption to plasticware during traditional tip-based serial dilutions can significantly impact potency determination and confound the understanding of structure-activity relationships used for lead identification and lead optimization phases of drug discovery. Here methods for automated acoustic non-contact serial dilution of peptide samples are described.
